چکیده

Photodynamic therapy (PDT) is an innovative and perspective antineoplastic therapy. Tetra-α-(4-carboxyphenoxy) phthalocyanine zinc (TαPcZn)-mediated PDT (TαPcZn-PDT) has shown antitumor activity in some tumor cells, but the manner which caspase-1 involved regulation of apoptosis pyroptosis TαPcZn-PDT-treated breast cancer MCF-7 cells unclear. Therefore, effects TαPcZn-PDT on cytotoxicity, cell viability, apoptosis, pyroptosis, cellular reactive oxygen species (ROS), mitochondrial membrane potential (ΔΨm), caspase-1, caspase-3, nuclear transcription factor-κB (NFκB) was firstly examined present study. The findings demonstrated that resulted increase cytotoxicity percentage apoptotic pyroptotic reduction viability ΔΨm, production ROS activation caspase-3 NFκB cells. Furthermore, results also revealed siRNA-targeting (siRNA-caspase-1) attenuated effect Taken together, we conclude regulates induced by

Crosstalk between p38 MAPK and caspase-9 regulates mitochondria-mediated apoptosis induced by tetra-α-(4-carboxyphenoxy) phthalocyanine zinc photodynamic therapy in LoVo cells

Photodynamic therapy (PDT) is considered to be an advancing antitumor technology. PDT using hydrophilic/lipophilic tetra‑α-(4-carboxyphenoxy) phthalocyanine zinc (TαPcZn-PDT) has exhibited antitumor activity in Bel-7402 hepatocellular cancer cells. However, the manner in which p38 MAPK and caspase-9 are involved in the regulation of mitochondria-mediated apoptosis in the TαPcZn-PDT-treated LoVo...

Reconstitution of caspase 3 sensitizes MCF-7 breast cancer cells to doxorubicin- and etoposide-induced apoptosis.

MCF-7, a breast cancer-derived cell line, is deficient of caspase 3 and relatively insensitive to many chemotherapeutic agents. To study the association of caspase 3 deficiency and chemotherapeutic resistance, we reconstituted caspase 3 in MCF-7 cells and characterized their apoptotic response to doxorubicin and etoposide. Western blots demonstrated that caspase 3 was constitutively expressed i...

Capsaicin-induced apoptosis in human breast cancer MCF-7 cells through caspase-independent pathway.

Capsaicin (trans-8-methyl-N-vanillyl-6-nonenamide), a significant pungent ingredient in a variety of red peppers of the genus Capsicum, is a type of vanilloid. It has been shown to exert biological activities (anticarcinogenic, antimutagenic and chemopreventive) in many cancer cell lines. It was found that capsaicin induces dose-dependent growth inhibition of MCF-7 cells, which does not express...

P-195: Thymoquinone Increases Efficacy of Tamoxifen Induced Apoptosis in Human Breast Cancer MCF-7 Cells: In Vitro

Background: The objective of this study is to evaluate combined effect of Thymoquinone (The main active component of black seeds) with Tamoxifen drug on apoptosis of human breast cancer MCF-7 cells (Noninvasive human breast cancer cell line, estrogen receptor positive). Materials and Methods: The human breast cancer MCF- 7 cells were treated with Tamoxifen (2 μM) alone or in combination with Th...
